Effects of Temperature on Enzyme Biology Introduction In order to understand the activity of enzymes at different temperatures the ability of the enzyme to function can be measured. This is important in many applications such as Polymerase Chain Reaction for forensics as well as genetics research where manipulation of temperature-dependent enzymes allows for replication of DNA segments. Bennett states, â€Å"when the energy – measured as heat – increases in a chemical reaction the energy increases the reaction speed and reaction rate. (1969) This happens until a threshold though in many biological processes where positive energy input no longer increases reaction speed due to the inhibition of other factors – in this experiment that factor is the degradation or denaturing of the protein complex that is the enzyme working to catalyze the reaction. One would make the hypothesis that for any given enzyme there is a minimum reaction rate which increases with tempera ture then at a maximum point begins to be less and less efficient as the enzyme form no longer works to catalyze the reactions with its given substrates.Materials and Methods After obtaining five clean test tubes and labeling them in order one through five, each were marked at one centimeter and at four centimeters from the bottom with a permanent marker. Next, the potato juice/catalase extract is added to each tube to the one centimeter mark. The following portion of the experiment was in segments for each tube. Tube one was placed in an ice-bath by adding ice and water in a 250ml beaker. Tube two was placed in the tube rack on the lab bench in a room temperature environment.Tube three was placed in a water bath at 50 degrees celsius, and tube four was set in the 70 degree celsius water bath. Tube five was placed in a boiling water bath of a beaker on the hot plate while wearing goggles for protection. Each tube was then left in the conditional environment for ten minutes. The temp erature of the boiling water, room temperature, and ice bath were measured by thermometer. After the ten minutes I used the thermometer to measure the temperatures of the room, ice, and boiling water. Then, after adding water, waited 30 seconds until the foaming in the tube, stopped.The measured the amount of foam was recorded. Results After all the appropriate steps were taken, a table was created and I recorded each temperature that was in the tube and foam height as well. The temperate and the foam height were both recorded to see the resemblance and results concluding the temperature effect on enzymes. An example of my results are recreated as seen below (numbers and titles are accurate and the same as lab test) As shown in the table A, temperature and foam height is represented by different color lines on the graph. The purple line, represents temperature. The blue line represents foam height (mm).As the temperature rose, the heigh of the foam decreased. This did not support my hypothesis. My hypothesis clearly stated,† If the temperature is really high, then the foam will rise highly. 50 degrees will be have the most amount of foam. † This hypothesis was not supported because the highest amount of foam was found at 0 degrees. The enzymes reached optimum temperature, and then denaturing took place. In comparison to another lab, the same results occurred. Campbell Neil from students. cis stated, â€Å"The higher the concentration of substrate, the faster the reaction rate. † This made my hypothesis invalid. DiscussionAlthough my hypothesis was not correct, the lab results agree with previous literature on the effects of temperature on enzyme activity. According to Susan Gilbert, â€Å"Enzymes are sensitive to their environmental conditions. Up to a point, the rate of the reaction will increase as a function of temperature because the substrates will collide more frequently with the enzyme active site. At extremes of pH or temperature, e ither high or low, the native structure of the enzyme will be compromised, and the molecule will become inactive†(2009). Enzymes are very sensitive and do the opposite of what most chemical reactions in this case.According to the Worthington Biochemical Corporation, â€Å"A ten degree Centigrade rise in temperature will increase the activity of most enzymes by 50 to 100%† (2012). Gary Sinise puts all of these camera angles and colours into one to create lots of suspense and to create different atmospheres. At the start of the film the screen is black and then white writing appears on top of the black background. There is light appearing through the planks of wood that we then know is a freight train. The casts and directors names appear as light peering through the planks of wood. The peering of the light acts a luminous spotlight at a prison and as it scans the area through the planks of wood it is like a light searching for someone. Also the planks of wood act as a cell and the light is glancing through the cell. All of a sudden the camera turns to this man who is sitting in a shadow in the corner of the freight train with half of his face in the light and the other half in the shadow. This implies I think that we can see straight through his eyes and see what he is remembering that is so vibrant. After this the director takes us to a women in a red dress running across a field with bright yellow straw and blue sky and green grass. This event had happened when the man on the freight train had a flashback of a past event and because the director has made all of these colours vibrant and alive this meant that the man’s memory was very fresh and bold. One of the first camera angles we see is at the start of the film on the freight train of the man’s face. The camera angle is a close-up of the man’s face. The director does this to show this through the man’s facial expression. At the start of the film there is a women running a cross a field in a bright red ripped dress. The camera stays where it is and the women runs towards the camera and this acts as if the woman is running towards us out of the screen to us for help. Also when the dogs and horses are running after the two men and trying to catch them it looks as if they are running straight out of the screen towards us. This makes us more involved and interested in the film. At the start of the film Gary Sinise creates mood and suspense by using different sound effects. He uses different sound effects so that they are bold and stand out in one particular scene so this makes the scene more interesting and has more action. At the start of the film there is a scene where there are men on horses and men with dogs chasing two men. The dogs and horses and men have sound effects because the men are breathing heavily so this makes it seem they have been running for a while to catch the two men. The dogs are barking viciously and the horses are neighing and these sound effects make the scene more enjoyable to watch and makes you feel you’re their with the dogs, horses and the men. He creates moods by using slow music in one scene then in the next scene using fast music and also the volume of the music. Then as the dogs, men and the horses start to gain some ground on the men to try and catch them the music starts to get faster this makes suspense and whether to see if the men will ever get caught. All of a sudden the men jump into a river and hide and as the men get closer and closer to them the music gets faster and faster then all of a sudden the music stops and the horses, men and dogs go away. This scene creates a lot of suspense and tension. The director Gary Sinise creates different mood sand suspense by putting colours, sound effects, camera angles and music into one. All of these effects create different moods like tension when the dogs, horses and the men are running after the men. The camera angles like the close up and the point of view create different moods because of what the character is thinking and what their emotions are these precise times. The music creates moods and tension by using the pace of the music fast or slow or the pitch or volume of the music. In the scene where the men are being chased the music gets quicker and quicker and builds up suspense and tension because you don’t know if the men are going to be caught or not. Should abortion be legal? Do you believe in Pro-Life or Pro-Choice? What do you do when circumstances come about that is not of your control or wiliness? I believe that both pro-life and pro-choice viewpoints have valid arguments.  These are the questions that we will examine briefly and what has happen since it became legal in the Roe vs. Wade Supreme Court decision case of 1973. It is important to know all that is involved with abortion before you can make a rational choice. It is not whether it should be legal, but when it should be allowed. Abortion allows women to have the choice and can save thousands of lives of women that have tried to do it on their own. I feel that it should be legal, but only to an extent. That is what pro-choice is all about. Pro-choice is the position that also is known as pro-abortion In the United States and in China, where law requires sometimes abortion and it is also known as anti-abortion. The purpose of the pro-choice movement is to ensure that all choices remain legal. The movement argues that a human person cannot be proven, prior to the point of viability and the government does not have the right to impede a womans right to decide whether to continue a pregnancy.(Head, 2010 )   Ã‚   I do not believe in permitting abortions after ten to twelve weeks of pregnancy, because at that time is when a fetus can represent potential life.   Now do not get me wrong, I still think that we should dissuaded women from abortion, and that we should be taking more measures to prevent unwanted pregnancies. It is because of the Roe vs. Wade case of 1973, states that a woman and her doctor can decide when and if to abort a pregnancy but only during the first trimester.
